Standard Operating Procedure (SOP) for Herbarium Presser
1. Purpose:
To provide a standardized method for operating and maintaining the herbarium presser to ensure proper pressing and preservation of plant specimens for herbarium collections.
2. Scope:
This SOP applies to all personnel involved in using the herbarium presser in the laboratory or herbarium.
3. Responsibility:
- Herbarium Technicians: Operate the herbarium presser according to the SOP.
- Herbarium Manager: Ensure the SOP is followed and updated as needed.
4. Equipment and Materials:
- Herbarium Presser (manual or hydraulic)
- Plant specimens
- Newspaper or blotting paper
- Cardboard sheets or pressing boards
- Personal Protective Equipment (PPE): Lab coat, gloves
5. Procedure:
5.1 Pre-Operation:
- Ensure the herbarium presser is clean and free from any contaminants.
- Verify that the presser is in good working condition, with no signs of wear or damage.
- Prepare the plant specimens by ensuring they are clean and free from dirt or insects.
5.2 Preparing the Specimens:
- Arrange the plant specimens on newspaper or blotting paper, spreading them out to avoid overlapping.
- Place the prepared specimens between two cardboard sheets or pressing boards for stability.
- Ensure that each specimen is properly oriented and flattened.
5.3 Operating the Herbarium Presser:
- Open the herbarium presser and place the prepared specimens between the pressing plates or layers.
- Ensure that the specimens are evenly distributed and not crowded.
- Close the presser carefully, ensuring that the specimens are positioned correctly and no part of the plant is excessively folded or crumpled.
- Apply pressure gradually and evenly according to the presser’s specifications. For manual pressers, this may involve turning screws or levers. For hydraulic pressers, adjust the hydraulic pressure to the recommended level.
- Maintain the pressure for the recommended duration, usually 24-48 hours, or according to the herbarium’s protocol.
5.4 Post-Operation:
- Carefully open the herbarium presser and remove the pressed specimens.
- Inspect the specimens to ensure they are properly flattened and preserved.
- Replace any damaged or worn parts of the presser if necessary.
- Clean the herbarium presser according to the manufacturer’s instructions to remove any residual plant material or debris.
5.5 Maintenance:
- Regularly inspect the herbarium presser for signs of wear, damage, or malfunction.
- Perform routine maintenance according to the manufacturer’s guidelines.
- Replace any worn or damaged components as needed.
- Record maintenance activities in the equipment logbook.
6. Safety Measures:
- Always wear appropriate PPE when handling plant specimens and operating the herbarium presser.
- Handle specimens and equipment with care to avoid injury or damage.
- Follow all safety guidelines and protocols for working with herbarium equipment and materials.
7. Documentation:
- Record the date, time, and purpose of each use of the herbarium presser in the lab notebook or equipment logbook.
- Note any deviations from the SOP and report them to the herbarium manager.
- Document the condition of the specimens before and after pressing.
