Standard Operating Procedure (SOP) for Simple Microscope
1. Purpose:
To provide a standardized method for operating and maintaining a simple microscope to ensure accurate and reliable observations.
2. Scope:
This SOP applies to all personnel involved in the use of the simple microscope in the laboratory for viewing small objects or samples.
3. Responsibility:
- Laboratory Technicians: Operate the simple microscope as per the SOP.
- Lab Manager: Ensure the SOP is followed and updated as needed.
4. Equipment and Materials:
- Simple Microscope
- Prepared slides or samples
- Lens cleaning paper or microfiber cloth
- Immersion oil (if applicable)
- Personal Protective Equipment (PPE): Lab coat, gloves, safety goggles
5. Procedure:
5.1 Pre-Operation:
- Ensure the microscope is placed on a stable, flat surface free from vibrations.
- Check the microscope for cleanliness, and ensure that the lenses are free of dust and residues.
- Wear appropriate PPE before starting the procedure.
5.2 Setting Up the Microscope:
- Adjust the microscope to a comfortable working height and position.
- Ensure the light source (if available) is working correctly. Adjust the mirror or light source to provide optimal illumination.
5.3 Preparing the Slide:
- Prepare the sample on a clean slide, placing a coverslip over the sample if necessary.
- Position the slide on the stage, securing it with the stage clips.
5.4 Focusing the Microscope:
- Start with the lowest magnification lens. Look through the eyepiece and adjust the mirror or light source to provide even illumination.
- Use the coarse adjustment knob to bring the sample into rough focus.
- Once the sample is visible, use the fine adjustment knob to achieve a clear and sharp image.
- If higher magnification is needed, rotate the nosepiece to switch to a higher magnification lens and use the fine adjustment knob to refocus.
5.5 Viewing the Sample:
- Observe the sample through the eyepiece, adjusting the focus and illumination as needed.
- Use both eyes to view the sample if the microscope has binocular eyepieces.
- Move the slide gently to observe different areas of the sample.
5.6 Post-Operation:
- After observing the sample, rotate the nosepiece to the lowest magnification lens.
- Remove the slide from the stage and clean any residue from the lens and slide using lens cleaning paper or a microfiber cloth.
5.7 Maintenance:
- Regularly clean the lenses and other optical components using lens cleaning paper or a microfiber cloth.
- Inspect the microscope for any signs of wear or damage.
- Record maintenance activities in the equipment logbook.
6. Safety Measures:
- Handle slides and lenses with care to avoid damage.
- Ensure proper ventilation in the area where the microscope is used.
- Dispose of used slides and samples according to the laboratory’s waste disposal guidelines.
